Tough to answer without more info...

In basic terms I would say the V30 has a more pronounced midrange through mid highs - although not "shrill" to my ear.
The G12K has a more pronounced low to low mid range.
Both are great speakers and you are comparing two similar speakers in my opinion.

I personally prefer V30s for recording. I prefer G12K for live. They can both do either just fine.

May I ask why you arrived at any of those two specifically? It all comes down to what you want from your gear, really, and the V30 and K100 are in many ways eachothers opposites, actually, hence a lot like to combine the two. Then there are other manufacturers of speaker drivers thay may suit your needs better as well.

Good question -- I just happen to have those two speakers lying around, and was trying figure out which might be the best fit.

Speakers are really a personal thing and the best advice I can ever give is to try them. Fortunately you have both in your possession already, so it cost nothing but your time to try both.

I personally like the V30 best in a blend with another speaker and never much on it's own. I used G12K-100's as my main speaker for live work for a few years in my last band and really liked it. As of late it hasn't been what I need and I have sold many of them off. Both are good speakers nonetheless.

Well, why don't you just put them in there and give them a whirl - unless of course you can have them mounted in separate cabs so you can switch between them quicker than in turn having to screw them into place.

I agree with Jaded Faith and others that it's a very personal thing, speakers. Maybe you don't like either of them. I myself went through most Celestions out there before I found what I was looking for in Fane speakers (I run a multi-amp setup).

As a general guideline the K100 is like a bassier and slightly congested G12-80, which is pretty neutral and has a rather forgiving yet present midrange. Great for rhythm, goth, death, doom and other rather dark and downtuned stuff, I would say - or bluesy and a bit smokey leads perhaps. If your amp/ modules has lot of mids and on top then this would be my choice.

The V30 of current manufacture has a bit of a spotlit mid that may work well with a neutral or, with lots of distortion, darker amp and can have a rather woody vibe in the 'right' rig. Can be a bit buzzy and honky with the 'wrong' rig since the range spotlit is rather rough and focused at a region our ears are particularly sensitive to. For instance a PAF into a Marshall Plexi MV into V30s can be a bit brash and brassy, which I'm sure may do well for somebody into punk, thrash, blackmetal or in other ways favours getting noticed over elegance.

Personally, I find V 30's to be obnoxious sounding but I play blues rock with humbuckers and they have a lot of mids already. I use WGS British leads which are similar to the K100 's and both are more of a neutral mid speaker.
if I played more Strats and Teles the v30 's would probably work better for me.

Well, I just swapped out the stock 75 for an Elite 80 I had lying around. It sounds awesome! Very little beaming, smooth yet present, and a tight low end. You might want to check those out, and they could be found used for cheap.

Wasn't the hottest in my TM212x (replaced with a British v30 and an old 85), but the E80 is awesome in an rm50 combo.

It is so easy to change the speakers in the rm50. You should just try them both I tried the wgs version of the vintage 30' and the classic lead 80, also the ET65. My favorite is the classic lead. But I put it and the vintage 30 in a 2x12 cab and that is what I use now. The et65 is a great speaker as well but sounded better in a different amp I own. Look for cheap speakers on eBay and sell what you don't like. I think eBay has a bunch of wgs speakers right now that are pretty cheap.

I like the crunch/grid/growl of the Celestion G12K100, but the low end is still not very tight when used in a 1x12 Thiele cab, althoug it's one of the tightest Celestion's on the market.
As a long time EV200 user and V30 lover I was looking for a hybrid speaker. Mixing speakers in a 1x12 Thiele (stereo setup) is not an option ;-)
Tried the Eminence V12, Tonker, Delta Pro, Fane Axa Studio 12L, Fane Medusa 150. The EV beats live all other speakers...
The G12K100 comes close but the real winner in mixing EV and Celestion in 1 speaker is the Eminence Super V. My 0,02 ct
